Two differential binding mechanisms of FG-nucleoporins and nuclear transport receptors

Summary Phenylalanine-glycine-rich nucleoporins (FG-Nups) are intrinsically disordered proteins, constituting the selective barrier of the nuclear pore complex (NPC). Previous studies showed that nuclear transport receptors (NTRs) were found to interact with FG-Nups by forming an “archetypal-fuzzy” complex through the rapid formation and breakage of interactions with many individual FG motifs. Here, we use single-molecule studies combined with atomistic simulations to show that, in sharp contrast, FG-Nup214 undergoes a coupled reconfiguration-binding mechanism when interacting with the export receptor CRM1. In this thesis, we studied the interactions between an anionic polysaccharide (pectin) and monovalent cation (Na+) and divalent cations (Ca2+, Zn2+, Ba2+, Mg2+) in dilute regime (c Ca2+ > Zn2+ > Mg2+; this may be related to the affinity between the water molecules from the coordination sphere and the cation. Indeed, the affinity of the cation for water molecules increases in the reverse order: Ba2+ < Ca2+ < Zn2+ < Mg2+. Finally, we have used the three polysaccharides (PGA, LMP and ALMP - amidated low methoxyl pectin) in association with calcium ions to produce microparticles containing rutin to target drug release in the intestine.
